The project will create human subject-specific induced pluripotent stem cells from deidentified repository specimens, use these to prepare human neuron cultures, and test the effect of alcohol treatment on gene expression, neuronal development, and neurophysiology. Gene editing and/or gene variant selection will incorporate human genetic information linked with alcohol abuset. The goal is to identify neuronal mechanisms underlying alcohol usage disorders.

Minimum Education and Experience

A Ph.D. and at least one year of relevant postdoctoral research experience is required. The successful candidate will have demonstrated experience in culture of human induced pluripotent stem cells and differentiation of these cells into electrophysiologically mature neurons. Experience in electrophyiology is required. Evidence of experience is first-author (or equivalent) publication in peer-reviewed research journals. Due to the highly collaborative nature of this project, excellent communication skills are required, as evidenced by authorship of first-author research journal articles and references from tenured faculty attesting to oral communication skills and ability to interact with other scientists.

Required Knowledge, Skills, and Abilities

The position will lead completion of an NIH-funded research project on alcohol use disorders (AUD). A Ph.D. and suitable postdoctoral experience is required to demonstrate effectiveness in this role. Background and knowledge in neuroscience, electrophysiology, alcohol research, and scientific communication skills are all required for the person to complete this assignment successfully.

Equipment Utilized

The project requires usage of BSL2 biosafety cabinets, fluorescent microscopy, electrophysiology equipment, and general molecular biology lab equipment (centrifuges, thermal cyclers, micropipets, etc.).

Physical Demands and Work Environment

The candidate must have sufficient manual dexterity to operate micropipets, fluorescent microscopes, and related equipment. The candidate must be cabable of working in a BSL2 biosafety cabinet for extended periods.

Overview

Prepare induced neurons from human induced pluripotent stem cells. Using lentiviral vectors, neuron-specific transcription factors drive reprogramming to stem cells to neurons. Immunocytochemistry and other methods will confirm identity of neurons. The candidate will have assistance from a technician. Perform experiments treating neuronal cultures with alcohol and evaluate changes in gene expression, electrophysiology, and differentiation markers. Perform gene editing on human induced pluripotent stem cells. To knock out or alter selected genes, the candidate will design reagents, introduce CRISPR/Cas9 components into cells, and screen for successfully edited cells.
